Varicella-Zoster Virus (VZV)

Varicella-zoster virus (VZV) is a member of the DNA virus family that plays a crucial role in triggering chickenpox and shingles. Chickenpox, one of the most prevalent diseases associated with this virus, is marked by the emergence of fluid-filled blisters on the skin, along with symptoms like itching and fever. VZV is a herpesvirus that exclusively infects humans, and is capable of reproducing in the lungs and leading to a range of symptoms. After the initial chickenpox infection, the virus enters a dormant state in the body and remains in the ganglion cells. VZV can reactivate when the body's immune system weakens, particularly with age or impaired function. Recombinant VZV proteins are frequently employed as antigens for in vitro diagnostic (IVD) procedures, aiding in the preparation of top-notch diagnostic agents used for identifying VZV-specific antigens or antibodies in bodily fluid samples from patients. This way facilitates the detection and screening of VZV infections.

Laboratory Tests

• Herpes scrapings. The relatively fresh herpes basal tissue is scraped off smeared and stained, and inclusion bodies can be seen within the multinucleated macrophages, indicating an association with the virus, which is a more specific manifestation.
• Virus isolation. Herpes fluid is inoculated into human dermal fibroblasts, and then the virus is isolated and finally visualized by electron microscopy, once the isolation is successful the diagnosis is confirmed.
• Immunologic testing. Antibodies can be detected by the complement binding test, which appears in the serum of chickenpox patients 1-4 days after diagnosis. Direct immune antibody staining or indirect fluorescent antibody method can also be used to check the antigen.
• Biotechnology. Detection of viral DNA in respiratory epithelial cells or peripheral blood by PCR is used as a means of early and rapid diagnosis.
